ANTI-INFLAMMATORY ACTIVITY OF A ZINC-CONTAINING COMPOSITE IN THE PRESENCE OF SIMULATED ULCERATIVE COLITIS

Introduction. Zinc is an essential nutrient that has a multidirectional activity during immune responses, including anti-inflammatory effects. An insufficient range of zinc-containing medicines and their low activity in the treatment of colitis justifies the need to design more effective zinc-based drugs. A zinc-containing biocomposite that is a hydrophobic rectal ointment has been designed. The pharmacologically active components of the ointment are a dihydroquercetin zinc-containing metal complex derivative containing 8.5-9.2% of zinc ions, as well as the bark wax of Siberian larch (Larix sibirica) and Gmelina. Objective: to evaluate anti-inflammatory properties of the designed zinc-containing biocomposite in the presence of simulated ulcerative colitis Material and methods. An experiment was carried out on 60 adult male Wistar rats. Ulcerative colitis was induced by an original procedure («Procedure for simulating ulcerative colitis», Russian Federation’s patent for the invention No. RU 2604044 dated December 10, 2016, Bulletin No. 34). Red blood cell and white blood cell counts and blood cell morphology differentials were determined by conventional methods. Results. The manifestation of erosive and ulcerative lesions of the colon mucosa in rats receiving the designed biocomposite was less marked than that in the control animal group, which was shown by a significantly less pronounced inflammatory reaction and no decreased red blood cell counts at all stages of the experiment. Conclusion. Rectal administration of the zinc-containing biocomposite in animals with simulated ulcerative colitis limits inflammation, which is confirmed by a minimum weight reduction by 10 days of the experiment and by the normalization of venous blood leukocyte and segmented neutrophil counts in progressive disorders in the comparison groups.
