DEVELOPMENT OF A PROCEDURE FOR QUANTIFICATION OF POLYSACCHARIDES IN HAWKWEED OXTONGUE (PICRIS HIERACIOIDES) HERB

Introduction. Hawkweed oxtongue (Picris hieracioides L.) in the Aster family (Asteraceae) is widespread in the middle belt of the European part of Russia. One of the groups of the active substances of hawkweed oxtongue is polysaccharides, which requires the development of a procedure for their quantification for the standardization of raw materials. Objective: to develop a procedure for gravimetric determination of polysaccharides and to validate the procedure. Material and methods. The investigation object was the herb of hawkweed oxtongue stored in its flowering period in the Central Black Earth regions in 2015–2017. The basis for quantification of polysaccharides is a gravimetric method. Results. The optimal extraction and precipitation conditions for polysaccharides were studied: these were as follows: the fineness of raw material grinding was 2 mm; extraction time was 120 min (four times for 30 min for each step); the ratio of water extraction to 96% ethyl alcohol was 1:3. The developed procedure was validated. The total content of polysaccharides in the examined samples ranged from 7.81 to 10.77%. Conclusion. The procedure has been developed for gravimetric determination of the amount of polysaccharides in the hawkweed oxtongue herb and validated by the following parameters: linearity, repeatability, and reproducibility.
