CHANGES IN THE LEVELS OF SOLUBLE PROTEIN AND FREE AMINO ACIDS IN TWO SPHAGNUM MOSS SPECIES

Introduction. The representatives of the bryoflora have been biochemically unstudied so far. In modern medicine, sphagnum mosses are limited in their use as wound dressing and antiseptic materials although there is evidence that the range of their application can be extended. Objective: to determine the content of soluble protein and to study the qualitative and quantitative composition of free amino acids in two species of sphagnum mosses collected in their different growing periods. Materials and methods. The plant material (brown sphagnum (Sphagnum fuscum), Baltic sphagnum (Sphagnum balticum)) was collected on the Chagino Swamp in the Tomsk Region in May to October 2010. Soluble protein was spectrophotometrically determined by the method described by Bradford. Free amino acids were quantified using the ninhydrin test after extraction with ethyl alcohol. Their qualitative composition was determined by paper chromatography. Results. The content of soluble protein and the qualitative and quantitative composition of free amino acids were studied in the test sphagnum species collected in different months of the growing season. It was demonstrated that the highest content of free amino acids in and their diversity of both moss species were observed in May and fell during the growing period while the level of soluble protein showed fluctuations around the mean value with a tendency to peak in July and October and to minimize in September. The qualitative composition of amino acids was determined in different months of collecting moss samples. The chromatograms displayed 3 to 10 individual spots of amino acids in relation to the month of material collection. Conclusion. The qualitative and quantitative composition of free amino acids and protein was determined in two sphagnum moss species – Sphagnum fuscum and Sphagnum balticum. Essential amino acids were found. The findings may be useful in designing new preparations based on the sphagnum moss tussock.
